Henry was 24 years old when South Carolinians fired the first shots on Fort Sumter, near Charleston, South Carolina, marking the official beginning of the War Between the States in April 14, 1861. A few weeks later, May 9, 1861, Henry Hanford Chamberlin, became the legal guardian for his minor sister, Sophronia Georgetta Chamberlin, Clark County Probate Court located in Springfield, Ohio in case no. A2754, Volume O, Page 36. Georgetta was 13 years of age. The same day Henry Hanford Chamberlin became legal guardian of his brother, James McPherson Chamberlin, age 18. Henry's father had died 10 years earlier.

On 12 Jun 1873, the Montgomery County Express, a Red Oak Junction newspaper reported, “The First National Bank of Red Oak is now fully organized with the following officers and directors: H.N. Moore, President; H. H. Chamberlin, cashier, Directors , Col. Alfred Hebard, Wayne Sennett, P.P. Johnson, H. A. McFartrich, S. D. Rymarsons, Henry Hanford Chamberlin, and H. N. Moore. Organized with a capital of $50,000.00.”
On June 26, 1873, the Montgomery County Express, a local newspaper reported, “This community was somewhat startled Saturday last by the announcement that H. H. Chamberlin of our city, had suddenly died of paralysis at Oskaloosa. Mr. Chamberlin was a prominent citizen of Red Oak, a noble hearted, enterprising man, and one of the largest property holders in the city; at the time of his death he was the cashier of the First National Bank, also a member of the Common Council of Red Oak"
