William and Charlotte had four children:
#William Cavendish, 5th Duke of Devonshire (1748–1811);#[[Cavendish-19|Dorothy Cavendish]] (27 Aug 1750 – 3 Jun 1794) m. PM William Cavendish-Bentinck, 3rd Duke of Portland, through her, the 4th Duke of Devonshire is the great-great-great-great grandfather of Queen Elizabeth II;
#Lord Richard Cavendish (1752–1781);#[[Cavendish-62 | George Augustus Henry Cavendish]], 1st Earl of Burlington, 2nd creation (1754–1834). Lord Burlington's grandson, the 2nd Earl of Burlington, would later inherit the Devonshire dukedom as 7th Duke of Devonshire.
William was elected as MP for Derbyshire in 1741. In June 1751, William took a seat in the Lords as Baron Cavendish and was soon after appointed to the Privy Council.
William's father died in December 1755 and he acceded to the dukedom.
William Cavendish Duke of Devonshire died on 2 October 1764, at Spa, the Austrian Netherlands (now Belgium).
