John was born around 1550 in Norwich, Norfolk, England, the son of Thomas Pettus, esq. (d. 1598) and his wife Christian Dethick.

John represented Norwich as one of two Members of Parliament having been elected on 27 February 1604/5 along with Sir Henry Hobart. He served until 1606.

He was knighted 29 June 1607 by King James I.

However, the engraving on his sumptuous family tomb at St Simon and St Jude reads::Here Under resteth the body of Sir John Pettus, Knighte, Cittizen and Alderman of Norwich who aged 64 departed this life the nynthe of Aprill 1613
John's very extensive Will was written on 10 January in the 11th year of King James (which converts to 10 January 1613/14). It was proved at the Prerogative Court of Canterbury in London by his second son Thomas (his first son Sir Augustine Pettus having died in 1613) with a probate date of 30 May 1614.
